DONATI’S LAB RESEARCH PROJECTS Applying ICP-MS for the Early Diagnosis of Osteoporosis

چکیده

The determination of chemical imbalances typically present at the onset of some aggressive diseases may be used for early diagnosis, which could reduce mortality and contribute for more efficient treatments. An interesting approach for nutritional and toxicological studies is the determination of minerals in fingernails and toenails. The concentrations of certain elements in these samples can be significantly different in healthy or sick individuals, and some works have demonstrated the efficiency of using nails as biological markers. More often than not, these elemental concentration changes are very subtle, especially at the onset of diseases. Therefore, a sensitive method is required to detect such small changes and contribute to diseases diagnostics. In this project, we take advantage of ICP-MS’ high sensitivity to identify subtle changes in the body’s mineral balance caused by osteoporosis. In collaboration with our colleagues at the Wake Forest Medical School, and Dr. Keith Levine with the Research Triangle Institute, we determine and compare the levels of different elements in fingernails and toenails of healthy individuals and patients with osteoporosis. We look for elemental signatures that could represent a less expensive, noninvasive method to identify at-risk individuals during the initial phases of osteoporosis.

The performance of single and multi-collector ICP-MS instruments for fast and reliable 34S/32S isotope ratio measurements†

The performance and validation characteristics of different single collector inductively coupled plasma mass spectrometers based on different technical principles (ICP-SFMS, ICP-QMS in reaction and collision modes, and ICP-MS/MS) were evaluated in comparison to the performance of MC ICP-MS for fast and reliable S isotope ratio measurements. The validation included the determination of LOD, BEC,...

Determination of Chromium, Selenium, and Molybdenum in Nutritional Products by ICP-MS Authors

An ICP-MS validated method for the rapid determination of chromium (Cr), selenium (Se), and molybdenum (Mo) in infant formula and adult nutritional products was successfully transferred from the Agilent 7500cx ICP-MS to the Agilent 7700x ICP-MS, and from lab 1 to lab 2, respectively. The key advantage of the method is that all samples can be analyzed using a single-cell gas mode (helium mode) t...

The value of lab findings in early diagnosis of acute mesenteric ischemia

Objective: Acute mesenteric ischemia (IMA) is a vascular emergency with broad variability of clinical presentations and non-specific laboratory findings. Therefore, there is a significant need for reliable serological markers of ischemia. Various laboratory studies may be performed for suspected AMI, but in general, such studies will not establish the diagnosis.Me...

Imaging of metals and metal-containing species in biological tissues and on gels by laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS): A new analytical strategy for applications in life sciences*

Laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) has become established as a very efficient and sensitive trace, ultratrace, and surface analytical technique in the life sciences. We have developed a new analytical imaging technique using LA-ICP-MS to study element distribution in biological tissues. Nowadays, imaging LA-ICPMS using double-focusing sector field (LA-ICP-SF...

Development and Validation of a GC-FID Method for Diagnosis of Methylmalonic Acidemia

Background: Urinary organic acids are water-soluble intermediates and end products of the metabolism of amino acids, carbohydrates, lipids, and a number of other metabolic processes. In the hereditary diseases known as organic acidurias, an enzyme or co-factor defect in a metabolic pathway leads to the accumulation and increased excretion of one or more of these acidic metabolites. Gas chromato...
